Genetic structure at different spatial scales in the pearl oyster (Pinctada margaritifera cumingii) in French Polynesian lagoons: beware of sampling strategy and genetic patchiness ArchiMer

In order to study further the genetic structure of the pearl oyster Pinctada margaritifera in French Polynesia with a special consideration for the sampling scale, we analyzed or re-analyzed sets of data based on nuclear DNA markers obtained at different spatial scales. At a large scale (several 1,000 km), the remote Marquesas Islands were confirmed to be significantly differentiated from Tuamotu-Gambier and Society archipelagos, with a marked difference however for the two main islands that are different from each other. At a medium scale (several 10 to several 100 km), overall homogeneity was observed within and between these two archipelagos, with some exceptions. Transmission expérimentale du virus de type herpes chez l'huître creuse, Crassostrea gigas ArchiMer

Au cours de l'été 1991, d'importantes mortalités de larves d'huître creuse, Crassostrea gigas, ont été observées dans des écloseries, simultanément en France (Comps et Cochennec, 1993) et en Nouvelle-Zélande (Hine & al., 1992). Les examens histologiques ainsi que des examens réalisés en microscopie électronique à transmission d'échantillons de larves présentant des mortalités ont permis de détecter la présence de particules virales associées à des lésions cellulaires. D'après la morphologie, la taille, les localisations intranucléaires et cytoplasmiques du virus, il est possible d'envisager son appartenance à la famille des Herpesviridae. Viruses infecting marine molluscs ArchiMer

Although a wide range of viruses have been reported in marine molluscs, most of these reports rely on ultrastructural examination and few of these viruses have been fully characterized. The lack of marine mollusc cell lines restricts virus isolation capacities and subsequent characterization works. Our current knowledge is mostly restricted to viruses affecting farmed species such as oysters Crassostrea gigas, abalone Haliotis diversicolor supertexta or the scallop Chlamys farreri. French scallops: A new host for ostreid herpesvirus-1 ArchiMer

Sporadic high mortalities were reported among larval French scallops (Pecten maximus). Electron microscopy of moribund larvae revealed particles with the characteristics of a herpesvirus in association with cellular lesions. PCR and DNA sequencing showed that the virus is a variant of ostreid herpesvirus-1 that has already been described in clams and oysters. This is the first description of a herpesvirus infection of a scallop species. The virus was transmitted successfully from an extract of infected scallop larvae to uninfected scallop or oyster (Crassostrea, gigas) larvae, demonstrating that it is able to infect both species. Evidence for interspecies transmission of oyster herpesvirus in marine bivalves ArchiMer

Since 1991, numerous herpesvirus infections associated with high mortality have been reported around the world in various marine bivalve species. In order to determine whether these infections are due to ostreid herpesvirus-l (OsHV1), a previously characterized pathogen of the Japanese oyster (Crassostrea gigas), PCR analysis was carried out on 30 samples of larvae collected from four bivalve species (C. gigas, Ostrea edulis, Ruditapes decussatus and Ruditapes philippinarum), most exhibiting mortality prior to collection. Molecular cloning and seasonal expression of oyster glycogen phosphorylase and glycogen synthase genes ArchiMer

To investigate the control at the mRNA level of glycogen metabolism in the cupped oyster Crassostrea gigas, we report in the present paper the cloning and characterization of glycogen phosphorylase and synthase cDNAs (Cg-GPH and Cg-GYS, respectively, transcripts of main enzymes for glycogen use and storage), and their first expression profiles depending on oyster tissues and seasons. A strong expression of both genes was observed in the labial palps and the gonad in accordance with specific cells located in both tissues and ability to store glucose. Among the different condition indices, some of them were compared on a population of the japanese oyster, Crassostrea gigas. The follow-up of experimental racks was performed monthly during four years. The use of dry weight in condition indices gave a more precise and more reliable representation for the condition of the oysters than the fresh weight. Early gametogenesis in the Pacific oyster: new insights using stem cell and mitotic markers ArchiMer

While our knowledge of bivalve gametogenesis has progressed in recent times, more molecular markers are needed in order to develop tissue imaging. Here, we identified stem cell and mitotic markers to further characterize oyster early gametogenesis, mainly through immunofluorescence microscopy. Intense alkaline phosphatase activity, a non-specific marker for stem cells, was detected on the outer edge of the gonad ducts at the post-spawning stage, suggesting an abundance of undifferentiated cells very early during the sexual cycle. Si les chimistes semblent satisfaits de leurs techniques de conservation, les biologistes quant à eux sont indécis du fait que l'action toxique se modifie au cours du temps et selon la technique employée (réfrigération, congélation, lyophilisation, conservation en azote liquide à -196°C). Le présent rapport constitue une étude préliminaire qui, si elle est poursuivie, pourrait permettre de déterminer le mode de conservation le plus approprié pour chaque type d'échantillons.

Arachidonic acid (20:4n-6, ArA) and its eicosanoid metabolites have been demonstrated to be implicated in immune functions of vertebrates, fish, and insects. Thus, the aim of this study was to assess the impact of ArA supplementation on the FA composition and hemocyte parameters of oysters Crassostrea gigas. Oyster dietary conditioning consisted of direct addition of ArA solutions at a dose of 0, 0.25, or 0.41 mu g ArA per mL of seawater into tanks in the presence or absence of T-Iso algae. The impact of diets upon the fatty acid composition of haemocyte polar lipids and consequently upon immune parameters has been tested in the oyster Crassostrea gigas and the clam Ruditapes philippinarum. Temperature triggers marine diseases by changing host susceptibility and pathogen virulence. Oyster mortalities associated with the Ostreid herpesvirus type 1 (OsHV-1) have occurred seasonally in Europe when the seawater temperature range reaches 16–24 °C. Here we assess how temperature modulates oyster susceptibility to OsHV-1 and pathogen virulence. Polyploidy in the Manila clam Ruditapes philippinarum. II. Chemical induction of tetraploid embryos ArchiMer

Tetraploidy was induced in embryos of Ruditapes philippinarum by treating the eggs with cytochalasin B. (CB). Two treatment times were found to be effective in inducing tetraplidy : 0 to 10 minutes after insemination (before formation of the first polar body) and 45 minutes after insemination (just before the first cleavage). Treatments, started 5 and 45 minutes after insemination, produced 64.4 ± 7.8% and 28.3 ± 2.3% tetraploid embrios, respectively, after 6 hours (on the average over 3 experiments). However, no tetraploids were detected in 4-month-old spat. Sperm entry delayed by the CB could be involved in induction of tetraploidy by early treatment.
